文章采用表面离子共振技术,建立了以VP28重组蛋白为靶点的抗白斑综合征病毒药物筛选平台,并对40味中草药进行了针对VP28重组蛋白结合活性的研究。结果显示:40味中草药中有14味中草药对VP28重组蛋白有明显结合力,含有结合VP28重组蛋白的活性成分分别为诃子、连翘、大黄、黄柏、苏木、白芍、紫花地丁、地榆、夏枯草、金银花、迷迭香、大青叶、黄连、黄芩。其中诃子含有的活性成分结合力最强,KD值为1.601 × 10−9,黄连的活性成分含量最高,RU值为76.483。文章首次利用该项技术创建了水产动物病毒性疫病药物筛选平台,为今后对虾白斑综合征药物的开发提供了理论支撑和科学依据。This article uses surface plasmon resonance technology to establish an anti-white spot syndrome virus drug screening platform targeting VP28 recombinant protein and studies the binding activity of VP28 recombinant protein on 40 Chinese herbal medicines. Results: Among the 40 Chinese herbs, 14 herbs have a significant binding affinity to VP28 recombinant protein and contain active ingredients that bind to VP28 recombinant protein, including Terminalia chebula Retz., Forsythia suspensa (Thunb.) Vahl, Rheum palmatum L., Cortex Phellodendri chinensis, Biancaea sappan (L.) Tod., Cynanchum otophyllum Schneid., Viola phillipina, Sanguisorba officinalis L., Prunella vulgaris L., Lonicera japonica Thunb., Rosmarinus officinalis L., Clerodendrum cyrtophyllum Turcz., Coptis chinensis Franch., and Scutellaria baicalensis Georgi. Among them, the active ingredients in Terminalia chebula Retz. have the strongest binding force, KD = 1.601 × 10−9, while Coptis chinensis Franch. has the highest content of active ingredients, RU = 76.483.
